An Experimental Alzheimer’s Drug Shows Promise Targeting a Different Brain Protein, New Study Shows

An experimental drug might help slow early Alzheimer’s disease in a markedly different way than today’s treatments — by lowering levels of a brain protein called tau, researchers reported Tuesday.

Tau is one part of a toxic duo fueling Alzheimer’s but prior attempts to develop drugs that can target the protein have failed. Two Alzheimer’s drugs, lecanemab and donanemab, try to clear buildup of the better-known amyloid protein and can modestly slow cognitive decline.

The new findings suggest Biogen's diranersen did more than lower tau levels. The study of about 400 people found signs that it also slowed cognitive decline, in one small subset enough to be comparable to amyloid therapy, according to results presented at the Alzheimer's Association International Conference in London. Biogen is planning a larger study to try to prove the drug’s benefit.

“This is really quite promising if it were to hold up” in that next-step testing, said Jessica Langbaum of the Banner Alzheimer’s Institute in Phoenix, who wasn’t involved with Biogen’s study.

“This is early days,” cautioned Dr. Reisa Sperling of Mass General Brigham, who also wasn’t involved in the study. But “I think it will reinvigorate interest and investment in lots of tau mechanisms, and the field needs that.”

It’s one of multiple novel attempts to fight the mind-destroying disease, including a possible tau vaccine, an experimental heart drug that might do double-duty for some people at high risk of Alzheimer's, and ways to help medicines more easily get across the so-called blood-brain barrier.

It’s not clear exactly what causes Alzheimer’s, which affects more than 7 million Americans and tens of millions worldwide. That sticky amyloid protein starts building up to form plaques in the brain about two decades before symptoms appear. But amyloid alone isn’t enough to cause Alzheimer's.

Many scientists believe that amyloid buildup eventually triggers an abnormal form of tau to form tangles in neurons, setting off symptoms.

Diranersen is what’s called an antisense oligonucleotide that doesn't attack tau buildup but instead instructs a tau-producing gene to produce less.

“If you lower tau production, you are lowering the amount of the abnormal tau that needs to be cleared by the microglia, by the clearance mechanism in the brain. And so, you are enabling the normal clearance mechanism to have more capacity to clear the tau,” said Dr. Cath Mummery of University College London, who led the new study.

Today’s anti-amyloid drugs are given through the bloodstream via infusions or injections. Diranersen is injected into the fluid surrounding the spinal cord, a straighter path to the brain.

Biogen's tau drug missed a key study goal — but was still encouraging Biogen’s study included people with mild cognitive impairment or mild Alzheimer’s, randomly assigning them to different doses of diranersen or a placebo.

Back in May, Biogen and partner Ionis Pharmaceuticals announced that the lowest dose — given every six months — had the strongest effect. That was a counterintuitive surprise and meant the study didn't meet its planned goal of showing that higher doses brought greater benefits.

Still, scientists had been anxiously awaiting details about how much that twice-a-year spinal shot really helped. Five of six different brain tests showed diranersen recipients’ memory and other cognitive abilities still worsened but more slowly than those given dummy shots, Mummery said.

In one test of the lowest dose, that translated to a 26% reduction in cognitive decline — “approximately the same” change seen in earlier tests of amyloid drugs, she said.

Side effects included injection site pain and a temporary state of confusion that could appear a few days after the shot and last about a week, she said. But there were no signs of brain inflammation, which can affect recipients of anti-amyloid drugs.

Alzheimer's researchers also target tau in a broad new study

The University of California, San Francisco, last week opened a first-of-its-kind study known as the Alzheimer’s Tau Platform.

Funded by the National Institutes of Health, it will test a variety of experimental anti-tau therapies against and in combination with today’s amyloid treatments. First up is a vaccine called AADvac1 designed to train the immune system to recognize and fight a specific worrisome portion of the tau protein, said UCSF's Dr. Adam Boxer.

The “platform” approach will expand to locations around the country, allow addition of other tau drugs to test and include people with Alzheimer’s-related protein buildup who aren’t yet showing symptoms, he said.

Other studies hint at new ways of attacking Alzheimer's

Researchers told the Alzheimer’s meeting that an experimental cholesterol-lowering drug called obicetrapib might do more than help heart health. They're exploring if it also might lower buildup of Alzheimer's-related proteins in people who carry a genetic risk for the disease.

Why? That gene, called APOE4, also affects how the body processes cholesterol. Obicetrapib maker NewAmsterdam Pharma plans to begin a study soon to test if the drug's cholesterol effects also can mitigate the Alzheimer's risk in people carrying one or two copies of that gene.

Companies also are trying to get Alzheimer’s drugs into the brain faster and at higher volumes, by penetrating the protective lining meant to protect the brain from harm.

Denali Therapeutics' CEO Ryan Watts describes it as “hitching a ride” with iron that naturally gets into the brain. His company is pursuing drugs that target tau and amyloid using that “transport vehicle” technology.

From a fishing trawler in the Adriatic, Italian scientists gently release thousands of tiny octopuses in the hope of fighting an invasive crab species that has devastated local fisheries.

A team from the University of Bologna is betting that the common octopus can help contain the burgeoning population of blue crab -- a voracious crustacean that has ravaged shellfish production in the Adriatic.

"We chose the octopus because, in the wild, it is a formidable predator of crustaceans," Antonio Casalini, one of the researchers, told AFP.

Native to the North American Atlantic coast, blue crabs arrived in the Mediterranean decades ago, likely transported via ballast water from ships.

But their numbers have ballooned in recent years, with the government and fishing groups linking the boom to warming waters and climate change.

With sharp, blue-tinted claws that are particularly apt at prying open shells, they have wreaked havoc in Italy's northeast -- one of Europe's top clam producing regions.

Authorities in Bologna's Emilia-Romagna region said last year the species has caused almost 17 million euros ($20 million) in damages there since 2022.

Farming and fishing group Coldiretti has put the figure at about 200 million for the whole of Italy, with the northern Veneto region also badly affected.

"It eats everything including young clams lying on the seabed," said Davide Sanulli a fishermen and mussels farmer with thick eyebrows and grizzled hair, said of the crab.

In lagoon-like areas off the Po River delta "it has caused a massacre", added the 64-year-old, who is helping the university project.

Excellent swimmers and weighing up to one kilogram (2.2 pounds), the crabs have few natural predators -- something the researchers are hoping to change.

Launched last year and funded by the Emilia-Romagna region, the "Octo-Blu" project seeks to increase octopus numbers as a counterweight to the hard-shelled invaders.

The team breeds octopus hatchlings in white water tanks at a lab in the port town of Cesenatico.

When ready, the tiny invertebrates are scooped out by the thousands into a large plastic bag and taken out to sea, sometimes using Sanulli's rugged blue fishing boat.

The hatchlings are then released near underwater reefs where the group has laid cement-and-clay artificial dens to help the octopuses thrive.

Some 80,000 have been let out since June, an amount the research team is seeking to double by the end of August, hoping to see a few grow into fully developed octopuses.

"The idea is to establish a small community close to shore so that it can better combat the spread of the blue crab," said Casalini, an aquaculture technician.

The project has some clear drawbacks.

The common octopus is not prone to live in the sandy, warm waters typical of the lagoon areas where the blue crab is making the most damage, making its potential impact there negligible -- something the researchers are well aware of.

Other experts have suggested that seabass could be more effective at crab-fighting and the Bologna University team is also looking at eels, which are avid eaters of crustacean eggs, to complement the octopus's efforts.

Yet, if replicated, the pilot could still help limit blue crab numbers and the related impact on the Adriatic coast's marine environment, Casalini said.

"Blue crabs have an incredible reproductive potential and fertility rate," said Pietro Emmanuele, another University of Bologna researcher, noting a single crab lays millions of eggs, most of which hatch.

Although only some hatchlings reach adulthood, were an octopus to catch even a single female, it would make a difference, he said.

The floating tentacled predators pulse and swirl in hypnotic patterns in the dark museum space, lit primarily by images projected on the walls and the glowing light from the saltwater tanks.

The private Jellyfish Museum in Pompano Beach, Florida, just north of Fort Lauderdale, is believed to be the first museum in the United States dedicated solely to the translucent invertebrates, and one of the few such spaces in the world, according to the city and the museum’s founders.

It was created by Alex and Yana Yanovsky, who previously developed successful aquariums in Ukraine, including their first jellyfish museum in Kyiv. The couple left those facilities behind when they fled Ukraine following the 2022 Russian invasion, operations manager Jessica Eisenbarth said.

The museum, which opened this past spring, is the couple's first aquatic attraction since immigrating to the US, but they'd like to expand.

“When they left Ukraine, they moved to Florida, and then they immediately wanted to share their passion here,” Eisenbarth said.

Jellyfish have survived for over 500 million years, making them older than dinosaurs and even trees. Their ancient origins are part of their allure, along with their hypnotic, pulsing movements. They have no heart or brain, and are made up primarily of water, giving them a translucent look.

Yana Yanovsky came up with the idea for the jellyfish museum after noticing how popular the jellyfish were at their other aquariums.

“Jellyfish are some of the most resilient creatures on Earth. While they lack a brain, they maintain a simple nerve net that allows them to respond to their environment,” Yana said in a statement provided by the city. “Their movement is mesmerizing, almost meditative, and they play an important role in marine ecosystems.”

Monty Graham, director of the Smithsonian Environmental Research Center, said he’s not surprised someone made an aquarium dedicated specifically to jellyfish. He credits the Monterey Bay Aquarium in California with kicking off the popularity of jellyfish exhibits at aquariums all over the world. Monterey Bay Aquarium began displaying jellyfish in temporary exhibits in 1985 and opened a permanent gallery in 1996.

“Jellyfish are cool, they’re things that people don’t get to see up close unless they’re scuba diving,” Graham said. “So here was this opportunity, in a controlled environment, very beautiful settings. It clicked all the buttons and checked all the boxes, and it was a very good, positive thing.”

The nearly 10,000-square-foot (900 square meters) Jellyfish Museum houses roughly 20 unique species of jellyfish found in Florida waters and other parts of the world. Different species can be rotated through the museum's 21 tanks. Some current occupants include Japanese sea nettles, moon jellyfish, spotted jellies and upside-down jellyfish, the last of which can be found among Florida's mangrove swamps.

The jellyfish are acquired through licensed aquaculture partners, according to the city.

The space is designed to exude calm and tranquility, general manager Iryna Pelypkanych said. It was converted from a former Wells Fargo bank branch with about $100,000 in community redevelopment grants from the city of Pompano Beach. Guides teach people about the role jellyfish play in marine ecosystems and the importance of protecting them.

“The biggest idea here is to leave people with the feeling that they know something more about the population of jellyfish,” Pelypkanych said. “Because so far, we know around 4,000 different species that can be called jellyfish.”

A NASA spacecraft circling the moon is providing the sharpest views yet of the crater carved by a crashing SpaceX rocket.

The space agency released before-and-after photos of the impact area on Tuesday.

The Falcon rocket’s upper stage plowed into the moon at 5,400 mph (8,700 kph) on Aug. 5 after drifting through space for more than a year. It launched a pair of private moon landers with experiments and even a tiny rover in 2025 as part of NASA’s push to commercialize lunar exploration.

Based on these new photos by NASA’s Lunar Reconnaissance Orbiter, the fresh crater appears to be 60 feet (18 meters) across and less than 10 feet (3 meters) deep, according to scientists.

Dark and bright streaks are clearly visible emanating like butterfly wings from the crater, The Associated Press reported.

The darker lines represent excavated material that was close to the surface and shaped by eons of solar wind, cosmic rays and micrometeorite strikes. The brighter rays indicate fresh rocks and dirt that were hurled from farther down.

The spacecraft photographed the crater a week after the collision from 60 miles (96 kilometers) up, while zooming along at one mile per second. Flight controllers had to tilt the spacecraft so its cameras pointed toward the impact scene.

The orbiter completes a lunar polar orbit every two hours, as the moon rotates beneath. It took six days before the crash scene came into view.

South Korea’s Danuri spacecraft was first on the scene to beam back photos.

The Lunar Reconnaissance Orbiter has been orbiting the moon since 2009, serving as NASA's up-close eyes as the space agency works to return astronauts to the lunar surface.
